Description

Biophysical characterization of binary and ternary complex formations in proximity-inducing compounds is a key step in the iterative improvement of potency of Proteolysis Targeting Chimeras (PROTACs) and molecular glues. In order for us to have the ability to scrutinise the affinity and kinetics of binary and ternary complexes with challenging sample requirements (e.g. low affinity (1-10 uM) complexes, low protein availability, homoPROTACs etc) highly specialised systems are required. The technique will provide a readout of compound/PROTAC affinity while providing insight into additional biophysical parameters, all of which can be critical for compound optimization/prioritization. This tender aims to substantially increase the ability of biophysicists and chemists to assess PROTAC/molecular glue biophysics and will be complimentary to techniques currently available to the new Center for Targeted Protein Degradation (CeTPD) (including SPR, ITC, TR-FRET) while being able to probe interactions that other techniques cannot currently scrutinise. The new machine will be housed in the CeTPD.
